Zane'nin gerçek aile soyadı Tzanetakos'dur. İlk büyük rolünü Geleceğe Dönüş filmi ile kazandı. Billy Zane, Uluslararası ününü Dead Calm filmindeki psikopat Hughie Warriner rolü ile yakaladı. Zane'in 1989 yılında rol aldığı bir diğer yapım ise NBC televizyonu için çekilen The Case of the Hillside Stranglers oldu. 1999 yılından itibaren pek çok film ve televizyon yapımında rolle almaya başlayan Zane, 1997 yılında çekilen Titanic filmi ile adından söz ettirmeye başladı. Bu rolü ile çok sayıda ödülün sahibi oldu. 1998 yılında çekilen I Woke Up Early The Day I Died filmi ile de çok sayıda ödül kazanmayı başardı.
